Zoloft: Antidepressants

The antidepressant effect of Zoloft is presumed to be linked to its ability to inhibit the neuronal reuptake of serotonin. Zoloft has only very weak effects on norepinephrine and dopamine neuronal reuptake. At clinical doses, Zoloft blocks the uptake of serotonin into human platelets. In receptor binding studies, Zoloft has no significant affinity for adrenergic (alpha(1), alpha(2) and beta), cholinergic, GABA, dopaminergic, histaminergic, serotonergic (5-HT1A, 5-HT1B, 5-HT2) or benzodiazepine binding sites.

Switching from other antidepressants or antiobsessional agents - There is limited controlled experience regarding the optimal timing of switching from other antidepressants or antiobsessional agents to Zoloft. Care and prudent medical judgement should be exercised when switching, particularly from long-acting agents such as fluoxetine. The duration of washout period which should intervene before switching from one selective serotonin reuptake inhibitor (SSRI) to another has not been established.
The study, which appears in the Oct. 27 issue of the Journal of the American Medical Association, found that cognitive-behavior therapy was more effective at relieving the symptoms of obsessive-compulsive disorder than treatment solely with the antidepressant sertraline (Zoloft).
