Vincristine

Objective: Vincristine has been widely used in various chemotherapeutic protocols in oncology. The purpose of this study was to evaluate the effect of vincristine on the biodistribution of 99mTc-DMSA, 99mTc-GHA, and 99mTc-DTPA in Balb/c female mice. Methods: Vincristine (0.03 mg, 0.3 mL) was injected into female isogenic Balb/c mice (n = 15), in 3 doses over an interval of 96 h. The 99mTc-DMSA, 99mTc-GHA, or 99mTc-DTPA (7.4 MBq) was administered after the last dose of vincristine. After 0.5 h the animals were killed rapidly. The organs (pancreas, thyroid, brain, thymus, ovary, uterus, spleen, kidney, heart, stomach, lung, liver, bone, and lymph nodes) were isolated and the radioactivity in each organ was counted in a NaI(Tl) well counter. The percentage of radioactivity (%) in each was calculated and compared with the control group.

Vincristine must only be administered by individuals experienced in the use of this cancer chemotherapeutic agent. Vincristine must only be administered intravenously, that is, directly into a vein. Accidental administration of vincristine into the spinal cord fluid is a medical emergency that may result in death. Vincristine has a low therapeutic index. It is unlikely there will be therapeutic benefit without toxic side effects. Certain complications can only be managed by a physician experienced in the use of cancer chemotherapeutic agents.

Vincristine (brand name, Oncovin)... known as [L]eurocristine, is a vinca alkaloid from the Madagascar periwinkle (Catharanthus roseus, formerly Vinca rosea and hence its name). It is a mitotic inhibitor, and is used in cancer chemotherapy.

Vincristine may reduce immunity to infections. No immunizations should be given while on vincrsitne. Vincristine may lower your body's resistance to infection and increase the chances of getting the disease the immunization is designed to protect you from.

Vincristine absolutely must be given intravenously. Vincristine is highly irritating to the soft tissues and if it is not given intravenously, where the bloodstream rapidly carries it away and dilutes it within the body's blood volume, it will cause what is called a tissue slough. This means the soft tissues will die and fall away leaving a large sore. Unlike the tissue sloughs of Adriamycin, a vincristine slough will eventually heal but while it is present, it will require bandaging and be a source of discomfort.

Vincristine can cause chemical burns if it comes in contact with the skin. If your child does not have a central line (port-a-cath, Hickman catheter, or PICC), a new IV stick will be required each time the dose is given.
