Propoxyphene: Analgesic
built 655 days ago
Propoxyphene is a weak narcotic analgesic which acts through binding to opiate receptors to inhibit ascending pain pathways. Propoxyphene, as with other narcotic (opiate) analgesics, blocks pain perception in the cerebral cortex by binding to specific receptor molecules (opiate receptors) within the neuronal membranes of synapses. This binding results in a decreased synaptic chemical transmission throughout the CNS ... inhibiting the flow of pain sensations into the higher centers. Mu and kappa are the two subtypes of the opiate receptor which propoxyphene binds to to cause analgesia.

Propoxyphene is an analgesic structurally similar to methadone. Its metabolite, norpropoxyphene, is ... pharmacologically active. Toxic effects include nausea, vomiting, and progressive central nervous system depression. Toxicity is additive with ethanol. Toxicity can be neutralized by narcotic antagonists. Peak serum level occurs 2 hours postoral dose; half-life is 8-24 hours.

Propoxyphene, a mild synthetic analgesic, is almost as effective as codeine. In combination with acetaminophen, it offers relief for mild to moderate pain, such as that caused by a bad toothache or the aftermath of minor surgery.

Propoxyphene may cause gastrointestinal (GI) upset. Propoxyphene-containing products may be taken with food to reduce or prevent GI upset.1 A common side effect of narcotic analgesics is constipation.2 Increasing dietary fiber (especially vegetables and whole-grain foods) and water intake can ease constipation.

Propoxyphene is a schedule C-IV controlled substance. Propoxyphene is a synthetic opiate agonist. Structurally, propoxyphene is more similar to methadone than to morphine. Compared with codeine, propoxyphene is one-half to two-thirds as potent an analgesic.
