Advagraf(R) is a once-daily formulation of Prograf(R) (tacrolimus,twice-daily formulation), the leading immunosuppressive agentdeveloped by Astellas. Advagraf(R) is expected to improve long-termcompliance with its more convenient once-daily dosing option, and mayhave the potential to reduce the incidence of chronic rejection andgraft loss. A recent study in kidney transplant recipients was ableto show a statistically significant improvement in adherenceassociated with once-daily dosing compared with twice-daily dosingregimens(1).

The principal adverse reactions of Prograf are tremor, headache, diarrhoea, hypertension, nausea, and renal dysfunction. These occur with oral and intravenous administration of Prograf and may respond to a reduction in dosing. Diarrhoea was sometimes associated with other gastrointestinal complaints such as nausea and vomiting.

Space your doses of Prograf as evenly as you can throughout the day. For example, if you take Prograf twice a day, doses should be 12 hours apart. Ask your transplant nurse or pharmacist about a dosing schedule that best fits your lifestyle.

Prograf Missed Dose: Take the missed dose as soon as you remember. If it is almost time for your next dose, skip the missed dose and take the medicine at the next regularly scheduled time. Do not take extra medicine to make up the missed dose.
