Pergolide Mesylate, better known as Permax, is a drug that is used to treat the signs and symptoms of Parkinson’s disease. However, in December 2002 the Mayo Clinic released a report claiming that patients that take Permax have the possibility of developing heart valve disease. Permax shares many characteristics of the diet drug fen-phen which has ... been accused of leading to heart valve disease. The Mayo Clinic’s study was based on three cases in which elderly women took Permax everyday for 3 to 7 years. The women were diagnosed with heart valve disease which prevents the hearts valves from properly closing during the hearts pumping motions. Since its introduction in 1989 it is estimated that over 500,000 people have taken the drug.

Permax is a category B FDA pregnancy medicine. This means that it has not been made clear whether taking this medicine during pregnancy could harm a growing fetus or not. However, if you are pregnant, or if you think that you might become pregnant any time soon, you should ask for your personal healthcare provider’s approval before you start taking this medicine. It is not known if this medicine’s ingredients are able to pass into the patient’s breast milk... harming the nursing child. However, you must not start taking this medicine without your personal physician’s approval if you are currently breast feeding an infant.

Drugs like Permax, Pergolide and Dostinex are known as dopamine agonists. Because Parkinson's disease is a progressive neurological disorder resulting from the degeneration of neurons in a region of the brain that controls movement, it causes a shortage of the brain-signaling chemical known as dopamine. The dopamine shortage creates the involuntary movement impairments that characterize the disease. Dopamine agonists initially act by stimulating some of the dopamine receptors, which assists in the management of the signs and symptoms of the disease. However, they cause the dopamine receptors to become progressively less sensitive, thereby eventually increasing the symptoms.

In January, 2007, the New England Journal of Medicine published two new studies which showed that Permax and Dostinex (cabergoline) may cause heart valve problems. Dostinex is used to treat Parkinson's disease in Europe, but in the U.S. is approved only for treatment of hyperprolactinemic disorders. One of the studies showed that Permax users were five to seven times more likely to have leaky heart valves than those taking other Parkinson's medications. A second study showed that one-fourth of Parkinson's patients taking Permax had moderate to severe heart valve problems.
