Naproxen is part of the class of drugs known as nonsteroidal anti-inflammatory drugs, or NSAIDs. Manufactured by German drugmaker Bayer and sold under the brand name Aleve, as well as Roche Palo Altos Naprosyn, naproxen was first sold by prescription in 1976 and was approved for over-the-counter sales in 1994. The popular over-the-counter drug is used to treat a variety of ailments, including headaches, menstrual cramps, arthritis and gout. Amidst drug safety warnings and controversy, naproxen is the latest drug to be added to the list of questionable medicines.

The clearance of Naproxen is 0.13 mL/min/kg. Approximately 95% of the Naproxen from any dose is excreted in the urine, primarily as Naproxen (<1%), 6-0-desmethyl Naproxen (<1%) or their conjugates (66% to 92%). The plasma half-life of the Naproxen anion in humans ranges from 12 to 17 hours. The corresponding half-lives of both Naproxen's metabolites and conjugates are shorter than 12 hours, and their rates of excretion have been found to coincide closely with the rate of Naproxen disappearance from the plasma. Small amounts, 3% or less of the administered dose, are excreted in the feces. In patients with renal failure metabolites may accumulate (see WARNINGS, Renal Effects).

Naproxen belongs to the group of preparations, which refer to non-steroid anti-inflammatory means (NAIM). These preparations are often used for treatment of an arthritis. Application of NAIM may have serious consequences for health down to a lethal result in consequence of a gastric or intestinal bleeding. The bleeding may arise at any time without any warning symptoms. Some doctors consider, that for the people older than 70 years there is better to begin treatment by preparations of this group from half of usual therapeutic doze.

Naproxen is associated with several suspected or probable interactions that affect the action of other drugs. The following examples are the most common suspected interactions. Naproxen may increase the blood levels of lithium (Eskalith) by reducing the excretion of lithium by the kidneys. Increased levels of lithium may lead to lithium toxicity. Naproxen may reduce the blood pressure lowering effects of blood pressure medications. This may occur because prostaglandins play a role in the regulation of blood pressure.
