In Killer Instinct, there are ten playable characters ranging from men made of fire or ice, to skeletons, to jungle queens, to professional boxers, among others. Each character has a repertoire of special moves. Each character is ... capable of attacking opponents with "combos." Combos are punch and kick combinations which result in an opponent being hit three or more times without the opponent being able to hit back or block. Killer Instinct is one of the first games developed to rely heavily upon combos as a major portion of the action. Combos typically do great damage to opponents.

[Y]eah, Killer Instinct was pretty fun. Of course, it has some learning disabilities when compared to other 2D and 3D fighers; not the most technical of the bunch. But fun! And stupid! And fun!

Contrary to what some believe, Killer Instinct does not use a modified Midway T-Unit architecture. Instead of the TMS34010, an R4600 RISC processor was used for the main CPU. This game does use the DCS DSP for sound and music, though. It showcased Nintendo's promotion for the Nintendo 64 system as the ULTRA 64.

Killer Instinct introduces Charlie Fox, a tough new heroine who can very definitely take care of herself. Charlie makes a living of sorts teaching self-defence to women. It's a skill she picked up out of necessity having been kicked out of the army for reasons she prefers not to go into. So, when Susie Hollins is found dead after she mistakenly tussles with Charlie at the New Adelphi Club, Charlie knows it's only a matter of time before the police come calling. What they don't tell her is that the unlucky Hollins is the latest victim of a homicidal rapist stalking the local area.

In the distant future, a monopolistic and technological corporation called Ultratech organizes a fighting tournament known as "Killer Instinct". Many fighters answer the challenge to participate, including several of Ultratech's experimental warriors. To increase the tournament's challenge, Ultratech utilizes a technology that can create bridges between alternate dimensions, releasing a creature known as Eyedol. Eyedol is a two-headed ancient warrior that was imprisoned along with his bitter rival, Gargos. Some warriors enter the tournament to seek glory or escape. Other fighters are attempting to bring an end to Ultratech's corruption.
