Indomethacin-induced G1 arrest and apoptosis of human colorectal cancer (CRC) cells is associated with a dose-dependent decrease in ß-catenin protein levels. ß-catenin plays a pivotal role in the WNT signalling pathway and its expression is frequently dysregulated at early stages of colorectal carcinogenesis. The objective of this study was to investigate the effect of indomethacin on catenin expression and downstream WNT signalling events in human CRC cells. ß-catenin, -catenin and T-cell facter (TCF) target gene (cyclin D1, c-MYC and PPAR) expression was studied following indomethacin treatment of SW480 and HCT116 cells. Cyclin D1 was used as a model TCF target gene for analysis of ß-cateninTCF-4 DNA binding and trans-activation. Indomethacin treatment was associated with a specific decrease in ß-catenin (but not -catenin) expression.

Indomethacin is used to treat patent ductus arteriosus and, on a different dosing schedule, to prophylactically decrease the risk of intraventricular hemorrhage. PDA in infants with RDS complicates mechanical ventilation, and is associated with BPD, decreases in cerebral, mesenteric, and renal blood flow, and CHF. Indomethacin, by inhibiting prostaglandin synthesis, allows the ductus to constrict in a high proportion of treated infants. Reversible renal insufficiency often occurs and fluid status must be carefully watched. Because significant oliguria can occur, fluids may need to be restricted to prevent fluid overload and dilutional hyponatremia. Do not follow BUN/Cr during Indomethacin therapy as they predictably rise.
