Indomethacin: Effects
built 618 days ago
Indomethacin farnesil is a novel antiinflammatory prodrug of indomethacin designed to reduce the occurrence of side-effects by esterification of the carboxyl group on indomethacin with farnesol. Indomethacin farnesil is marketed as trade names Infree and Dialon. CAS: 85801-02-1
Indomethacin appears to have fewer side effects on the mother but potentially more serious effects on the fetus than other tocolytic medications used to treat preterm labor. 3 However, fetal side effects are very unlikely when treatment lasts fewer than 7 days. Indomethacin may cause:
The effects of Indomethacin during pregnancy have not been adequately studied. If you are pregnant or plan to become pregnant inform your doctor immediately. Indomethacin appears in breast milk and could affect a nursing infant. If this medication is essential to your health, your doctor may advise you to discontinue breastfeeding until your treatment with this medication is finished.
Indomethacin has been reported to diminish basal and CO2 stimulated cerebral blood flow in healthy volunteers following acute oral and intravenous administration. In one study after one week of treatment with orally administered indomethacin, this effect on basal cerebral blood flow had disappeared. The clinical significance of this effect has not been established.

Indomethacin-induced responses in eosinophils were similarly modulated by inhibitors of PI 3-kinase, p38 MAPK, and MEK but with two exceptions (Fig. 6). First, inhibition of p38 MAPK reduced both the potency and efficacy of indomethacin in assays of eosinophil shape change, whereas the p38 MAPK inhibitor suppressed eotaxin-induced eosinophil shape change potency alone. Second, the MEK inhibitor U-0126 caused an insignificant inhibition of eosinophil shape change induced by submaximal indomethacin concentrations and had no effect on indomethacin-induced up-regulation of eosinophil CD11b expression.

CHILDREN The safety and effectiveness of Indomethacin have not been established in children under 14 years of age. However, your doctor may decide that the benefits of this medication outweigh any potential risks.
