Giantism is the result of excessive growth hormone secretion that begins in young children or adolescents. It is a very rare disorder, usually resulting from a tumor of somatotropes. One of the most famous giants was a man named Robert Wadlow. He weighed 8.5 pounds at birth, but by 5 years of age was 105 pounds and 5 feet 4 inches tall. Robert reached an adult weight of 490 pounds and 8 feet 11 inches in height. He died at age 22.

Unfortunately, there are not yet any really long-term clinical studies on the use of growth hormone replacement therapy. Most of the longer studies in persons suffering from only age-related conditions have used bizarre dosing regimens for HGH. This was understandable in the early studies, when the proper dosage in humans was unknown. It does NOT make sense that so many clinical studies continue to use such large doses. Overdosing on any hormone will inevitably lead to adverse effects. Most people using HGH to replace declining levels of growth hormone use one unit per day or less.

Forsythe, 69, is accused of twice selling vials of Bio-Tropin, an illegal human growth hormone sold in Israel, to an undercover Food and Drug Administration investigator in 2004. Special Agent John Zalinsky told Forsythe he was referred to him by a friend at his gym whom he was treating with HGH for anti-aging purposes.

Laboratory mice that have been genetically altered to produce human growth hormone grow to be 25-30 percent larger than normal mice - with much of that size difference coming from bigger bones, according to U-M researchers. The trade-off for bigger bones in adolescence appears to be poorer bone quality, increased brittleness and less resistance to fractures as mice age.

[I]n 2002, another study published in the Journal of the American Medical Association examined the effect of six months of HGH with and without hormone replacement therapy (HRT) on fifty-seven women and seventy-four men age 65 to 88. The women treated with growth hormone averaged a 2.2 pound increase in their muscle mass, if they took HRT alone they did even better. HRT and HGH gave them a 4.5 pound benefit. But with or without these hormones, there was no significant improvement in their strength or cardiovascular endurance (as measured with a treadmill). The men seemed to have a greater response to HGH, showing marginal improvement in their strength and endurance. The most concerning aspect of this study was the high rate of side effects.

In adults, growth hormone deficiency causes an increase in fat tissues and a decrease in muscle mass. In addition, growth hormone deficiency may negatively affect functioning of the heart (decreased cardiac output) or skeletal muscle (physical weakness). However, common (and perhaps major) complaints in adults may be lethargy and decreased sense of well-being that are frequently attributed to “old age”.
