Doxycycline (Dox), a derivative of tetracycline (Tc), is the recommended inducer for all of Clontech’s Tet Inducible Expression Systems. In the Tet-On® Advanced System, addition of Dox to the culture medium induces the rtTA-Advanced transactivator to bind to the Tet response element (TRE), this results in transcriptional activation of the gene of interest. Conversely, in the Tet-Off® Advanced System, removal of Dox from the culture medium induces binding of the tTA-Advanced transactivator, resulting in transcription of the gene of interest.

Doxycycline and other members of the tetracycline class of antibiotics are not generally approved for treating patients under 8 years old. These antibiotics can cause swelling of the brain and a condition called bulging fontanels (soft spot on head) in infants.

Doxycycline is less bound to the calcium in maternal milk which may lead to increased absorption compared to other tetracyclines. Only minimal amounts of doxycycline are excreted in human milk and the relative amount of tooth staining has been reported to be lower when compared to other tetracycline analogs. Nondose-related effects could include modification of bowel flora.
