Diclofenac sodium is a non-steroidal compound, a phenylacetic acid derivative, with analgesic, antipyretic and anti-inflammatory effects. Diclofenac sodium inhibits the biosynthesis and release of prostaglandins, which are known to be implicated in the pathogenesis of inflammation, pain and fever. ADCO-DICLOFENAC Tablets are enteric-coated so that absorption occurs in the gastrointestinal tract to give peak plasma concentrations approximately 2 hours after ingestion. There is at least 99% binding to plasma-proteins and excretion of metabolites is mainly in the urine.

Diclofenac Potassium Tablets, USP for oral administration, contain 50 mg of Diclofenac Potassium. In addition, each tablet contains the following inactive ingredients: anhydrous lactose, colloidal silicon dioxide, croscarmellose sodium, glyceryl triacetate, hypromellose, magnesium stearate, microcrystalline cellulose, polydextrose, polyethylene glycol, sodium lauryl sulfate and titanium dioxide.

Nursing mothers Diclofenac sodium has been found in the milk of nursing mothers. It is unlikely that misoprostol is excreted into milk since the drug is rapidly metabolized throughout the body. Excretion of the active metabolite misoprostol acid ; into milk is possible, but has not been studied. Misoprostol acid could cause significant diarrhea in nursing infants. Because of the potential for serious adverse reactions in nursing infants, ARTHROTEC is not recommended for use by nursing mothers. Labor and Delivery In rat studies with NSAIDs, as with other drugs known to inhibit prostaglandin synthesis, an increased incidence of dystocia, delayed parturition, and decreased pup survival occurred.

Diclofenac free acid was obtained from diclofenac sodium salt (Sigma Aldrich, Milano, Italy) by precipiting it with HCl. The obtained diclofenac free acid was filtered and dried at room temperature under a vacuum. β-cyclodextrin was kindly supplied by Roquette Co, Lestrem, France. Phosphate buffer solution (PBS) pH 7 was obtained from Carlo Erba Reagents (Rodano, Italy). High-performance liquid chromatography (HPLC)-grade methanol and propylene glycol were purchased from Sigma Aldrich.
