Smith Degradation of the Cell Wall-- The insoluble cell wall matrix, defined above as the product remaining after extraction with chloroform/methanol and SDS, was subjected to Smith degradation by a method previously described (6). Briefly, the cell wall was sonicated to a fine suspension in 10ml of 0.1M NaIO4 in 50mM sodium acetate buffer (pH 5.0) and kept in the dark for 5days. After extensive washing in water, the insoluble residue was reduced by stirring with 130mg of NaBH4 in 10ml of water for 5h. The reaction mixture was titrated to pH 5with acetic acid, pelleted, and treated with 1N HCl at room temperature overnight. The insoluble residue was washed with water and extracted with CHCl3. The CHCl3-soluble products were subjected to analysis by electrospray ionization (ESI) and matrix-assisted laser desorption/ionization (MALDI) mass spectroscopy.

image Eight Cell Wall and Resist Wall Flower Pots will be installed into EMCS, 4 on rotor A which will rotate at 1G and 4 on rotor B, which will not rotate and provide a microgravity environment. The crew will provide on-orbit support by checking the EMCS daily for nominal operation. Cultivation of these plants will last approximately 33 - 53 days. The Cell Wall and Resist Wall Flower Pots will be removed from EMCS and placed inside the Microgravity Sciences Glovebox (MSG), to prepare the plant material for harvesting. Crewmembers will divide, fix and store the samples using the Kennedy Space Center Fixation Tube (KFT), a system designed to contain plant or other small biological samples during flight and chemically fix and stain the tissue samples.

MYCAMINE inhibits an enzyme essential for fungal cell-wall synthesis and is fungicidal (lethal) for Candida. MYCAMINE can be used concomitantly with a variety of other drugs, including the HIV protease inhibitor ritonivir and the transplant medications cyclosporine and tacrolimus. Under the announced agreement, Roche will utilize its highly experienced hospital sales force to augment Fujisawa's promotion of MYCAMINE throughout the United States. Fujisawa will book sales and compensation to Roche will be based on product sales.

Rho1 is considered the master regulator of CWI signaling not only because it receives the major inputs from the cell surface but ... because it regulates a variety of outputs involved in cell wall biogenesis, actin organization, and polarized secretion (Fig. 1). Moreover, it seems likely that Rho1 coordinates these functions at the cell surface. It is localized to sites of polarized growth in a manner dependent on the actin cytoskeleton (16, 265, 359).

Researchers have known for some time that two enzymes, known as polygalacturonase and expansin, contribute individually to the ripening-related breakdown of the cell walls. (Enzymes are proteins that trigger and control chemical reactions.) The UC Davis research team wondered if these two cell wall enzymes might ... be responsible for the increased disease-susceptibility of ripening fruit. These diseases are responsible for substantial losses of high-quality harvested fruit during storage, shipping, marketing and consumer handling.
The binding of these cell wall components to their corresponding pattern recognition receptors triggers the macrophages to release various defense regulatory chemicals called cytokines (def), including IL-1, IL-6, IL-8, TNF-alpha, and PAF. The cytokines then bind to cytokine receptors on target cells and initiate inflammation and activate both the complement pathways (def) and the coagulation pathway (see Fig. 2).
